Melanotan 2 is a cyclic lactam analogue of the natural hormone alpha-MSH (cyclo[Nle4,Asp5,D-Phe7,Lys10]-alpha-MSH(4-10)) that binds four melanocortin receptors with high affinity: MC1R, MC3R, MC4R and MC5R. MC1R activation on melanocytes triggers melanogenesis, the eumelanin synthesis that darkens skin independently of UV light. MC4R activation in the hypothalamus and brainstem drives sexual motivation and erectile signalling (the same pathway later isolated in PT-141) while also suppressing appetite. MC3R contributes to energy homeostasis and MC5R to exocrine function. Because a single molecule engages the whole melanocortin system at once, Melanotan 2 produces the characteristic cluster of pigmentation, arousal and appetite effects seen across research studies.
